ABSTRACT

         Standardized karyotype and idiogram of the variable squirrel (Callosciurus finlaysoni bocourti) was studied at Khon Kaen University, Thailand. Blood samples were taken from two male and two female variable squirrels. After lymphocyte culture, the mitotic chromosome preparation was accomplished by the hypotonic-air-drying method and conventional Giemsa’s staining technique. The results showed that the number of diploid chromosomes was 2n=40 and the fundamental number (NF) was 78 in both male and female. The autosomes consisted of 4 large metacentric, 4 large submetacentric, 14 large acrocentric, 2 medium telocentric, 4 small metacentric, 6 small acrocentric and 4 small telocentric chromosomes. The X chromosome was a medium metacentric chromosome and the Y chromosome was a small metacentric chromosome. A pair of the short arm near centromere of chromosome pairs 14 showed clearly observable secondary constriction (satellite chromosomes). The chromosome marker of the variable squirrelis the chromosome pair 1 which is the largest submetacentric chromosome. The karyotype formula for the variable squirrel is as follows:
                  2n (40) = Lm4+Lsm4+La14+Mt2+Sm4+Sa6 +St4+ sex chromosomes

ABSTRACT

        Standardized karyotype and idiogram of the Indochinese ground squirrel (Menetes berdmorei (Blyth, 1849) at was studied at the Nakhon Ratchasima Zoo, Thailand. Blood samples were taken from two male and two female Indochinese ground squirrels. After lymphocyte culture, the mitotic chromosome preparation was accomplished by hypotonic-air-drying method and conventional Giemsa’s staining technique. The results showed that the number of diploid chromosome was 2n=38 and the fundamental number (NF) was 68 in both female and male. The autosomes consisted of 8 large metacentric, 14 large submetacentric, 2 medium metacentric, 4 small metacentric and 8 small telocentric chromosomes. The X chromosome was a medium metacentric chromosome and the Y chromosome was a small metacentric chromosome. The chromosome marker of the Indochinese ground squirrel is the chromosome pair 1 which is the largest metacentric chromosome. ABSTRACT

           Standardized karyotype and idiogram of the Thai native swamp buffalo (Bubalus bubalis) at Khon Kaen University, Thailand, were studied. Blood samples were taken from two male and two female swamp buffaloes. After lymphocyte culture, the mitotic chromosome preparation was accomplished by hypotonic-air-drying method. Conventional staining, G-banding, C-banding and NOR-banding techniques were applied to stain the chromosome. The results showed that diploid number of Thai native swamp buffalo was 2n=48, the fundamental numbers (NF) were 58 in both male and female. The types of autosomes were 4 large metacentric, 4 large acrocentric, 2 small submetacentric and 36 small telocentric chromosomes. The X chromosome was large telocentric chromosome and the Y chromosome was the small telocentric chromosome. From G-banding, each chromosome pair appears with distinctively differentiated. C-banding shown C-positive (dark band) on centromere of all telocentric chromosomes but others appeared as C-negative (light band). NOR banding exhibited 6 telocentric chromosome pairs. The karyotype formula of Thai native swamp buffalo was as follows:
                    2n (diploid) 48 = Lm4+ La4+Ssm2+St36 + sex chromosomes

The first chromosome characterization of the family Tragulidae 
(Artiodactyla) in Thailand by conventional staining

Abstract
         Karyotypes were studied from the family Tragulidae of Thailand, representing a single genus with two species namely; lesser Malay mouse-deer (Tragulus javanicus) and larger Malay mouse-deer (Tragulus napu). Blood samples were taken from the two species kept in Khoa Kheow Open Zoo, Chonburi province and Songkhla Zoo, Songkhla province, Thailand. After standard whole blood lymphocyte culture in presence of colchicine, the metaphase spreads were performed on microscopic slides and air-dried. Conventional Giemsa’s staining was applied to visualize chromosomes. The karyotype of lesser Malay mouse deer showed that diploid chromosome number was 2n=32 and fundamental numbers (NF) were 64 in both female and male. The autosomes consist of 6 large metacentric, 6 large submetacentric, 14 medium metacentric, 2 submetacentric and 2 small metacentric chromosomes. The X chromosome was a large submetacentric chromosome while the Y chromosome was a small metacentric chromosome. For our result, the first karyotypic study of T. napu, the larger Malay mouse-deer, the karyotype shows that diploid chromosome number was 2n=32, and NF were 64 in both female and male. The autosomes consist of 6 large metacentric, 6 large submetacentric, 12 medium metacentric, 2 medium submetacentric, 2 medium acrocentric and 2 small submetacentric chromosomes. The X chromosome was a large submetacentric chromosome while the Y chromosome was the smallest metacentric chromosome.

Abstract
        Cytogenetics of Thai brow-antlered deer (Cervus eldi siamensis) were studied in comparison with those of Thamin brow-antlered deer (Cervus eldi thamin). Blood samples were taken from the two subspecies kept in Khoa Kheow Open Zoo, Chonburi Province. After the standard whole blood lymphocyte were cultured in presence of colchicine, the metaphase spreads were performed on microscopic slides and air-dried. Conventional Giemsa’s staining were applied to stain chromosome. Thai and Thamin brow-antlered deers exhibited the same karyotype with diploid number of 2n = 58 (NF = 70) for females and 2n = 58 (NF = 71) for males. The types of autosome are 6 large metacentric, 6 large submetacentric, 8 large telocentric, 20 medium telocentric and 16 small telocentric chromosomes. In addition, satellites are clearly observed in terminal position on the short arm of a pair of chromosome 7. The X chromosome is the largest telocentric and the Y chromosome is the smallest metacentric chromosome. The karyotype formula of Thai and Thamin brow-antlered deer is as follows:
          2n (58) = Lm6+Lsm6+Lt8+Mt20+St16+sex chromosome

Abstract
        Cytogenetics of the pileated gibbon (Hylobates pileatus) at Nakhon Ratchasima Zoo, Thailand, was studied. Blood samples were taken from two female and two male gibbons. After lymphocyte culture, the mitotic chromosome preparation was done by hypotonic-fixation-air-drying method and conventional Giemsa’s staining. The results show that diploid chromosome number was 44 (2n=2x=44), and the fundamental number (NF) were 88 chromosomes in both female and male. The autosomes consist of 12 large metacentric, 6 medium metacentric, 2 medium submetacentric, 2 medium acrocentric, 12 small metacentric and 8 small submetacentric chromosomes. In addition, the chromosome 15 showed clearly observable satellite chromosomes. The X chromosome was a medium submetacentric chromosome and the Y chromosome was a tiny acrocentric chromosome. The karyotype formula for the pileated gibbon is as follows:
           2n (44) = Lm12+Mm6+Msm2+Ma2+Sm12+Ssm8+sex-chromosomes

บทคัดย่อ
การศึกษาคาริโอไปท์ของชะนีมือขาวจากสวนสัตว์นครราชสีมาทำขึ้นโดยการ เตรียมโครโมโซมจากการเพาะเลี้ยงเซลล์เม็ดเลือดขาวจากชะนีมือขาวเพศผู้ 2 ตัว และเพศเมีย 2 ตัว หลังจากนั้นเก็บเกี่ยวเซลล์ด้วยเทคนิคโคลชิซิน-ไฮโปโทนิค-ฟิกเซซั่น-แอร์ดรายอิง และย้อมสีโครโมโซมแบบธรรมดาด้วยสีจิมซ่า ผลการศึกษาพบว่าชะนีมือขาวมีจำนวนโครโมโซมดิพลอยด์เท่ากับ 44 แท่ง มีจำนวนโครโมโซมพื้นฐานเท่ากับ 88 ในเพศเมียและเพศผู้ โครโมโซมร่างกายประกอบด้วยโครโมโซมชนิดเมทาเซน ทริกขนาดใหญ่ 12 แท่ง เมทาเซนทริกขนาดกลาง 8 แท่ง ซับเมทาเซนทริกขนาดกลาง 4 แท่ง อะโครเซนทริกขนาดกลาง 2 แท่ง เมทาเซนทริกขนาดเล็ก 8 แท่ง และซับเมทาเซนทริกขนาดเล็ก 8 แท่ง แขนข้างยาวของโครโมโซมคู่ที่ 13 จัดเป็นแซทเทลไลท์โครโมโซม โครโมโซมเอ็กซ์เป็นชนิดซับเมทาเซนทริกขนาดกลาง และโครโมโซมวายเป็นชนิดอะโครเซนทริกขนาดเล็กมากที่สุด

Abstract
Karyotype of the white-handed gibbon (Hylobates lar) kept in Nakhonratchasima Zoo, was studied. Blood samples were taken from two female and two male gibbons. After lymphocyte culture, the mitotic chromosome preparation was done by air-drying method and conventional Giemsa’s staining. The results showed that diploid chromosome number was 2n=44 and the fundamental number (NF) were 88 chromosomes in both female and male. The autosomes consisted of 12 large metacentric, 8 medium metacentric, 4 medium submetacentric, 2 medium acrocentric, 8 small metacentric and 8 small submetacentric chromosomes. In addition, a pair of the long arm of chromosome 13 showed clearly observable satellite chromosomes. The X-chromosome was the medium submetacentric and the Y-chromosome was the smallest acrocentric chromosome.

ABSTRACT

          The study on chromosomal staining comparison of plant cells with natural dyes was carried out to compromise the use of expensive dyes. Dyes from black glutinous rice (Oryza sativa L.) and Lac (Laccifer lacca Kerr) were extracted using acetic acid, ethanol, butanol and hexane with the concentration levels of 30%, 45% and 60%, respectively. The pH was then adjusted from 1 to 7, the natural extracted dyes were used to stain the chromosomes of spider lily (Hymenocallis littoralis Salisb.) root cells, which were ongoing mitotic cell division, using the squash technique. The results showed that the natural extract dyes were capable of chromosome staining and cell division observing. Natural dyes which showed well-stained chromosome included 45% acetic acid-extracted black glutinous rice dye (pH 1–3), 45% butanol-extracted black glutinous rice dye (pH 1–3) and 60% ethanol-extracted Lac dye (pH 1–3). We also concluded that all other extracts have no significant quality as chromosomal staining indication.

ABSTRACT

         Standardized karyotype and idiogram of the Mehsani buffalo (Bubalus bubalis) at Burirum Livestock Research Station, Thailand, was studied. Blood samples were taken from two male and two female buffaloes. After standard whole blood lymphocytes were cultured at 37oC for 72 h in the presence of colchicine, the metaphase spreads were prepared on microscopic slides and air-dried. Conventional staining, GTG-banding, CBG-banding and Ag-NOR banding techniques were applied to stain the chromosomes. The results showed that the diploid chromosomes number of Mehsani buffaloes was 2n=50; the fundamental numbers (NF) were 60 in both male and female. The types of autosomes were four large metacentric, six large submetacentric, eight large telocentric, eight medium telocentric and 22 small telocentric chromosomes. The X chromosome was a large telocentric chromosome and the Y chromosome was a small telocentric chromosome. In GTG-banding, each chromosome pair appeared clearly differentiated. CBG-banding showed dark bands on the centromere of all telocentric chromosomes (autosome pairs 6-24 and the X chromosome) but light band on the others (autosome pairs 1-5 and the Y chromosome). Ag-NOR banding exhibited six position of autosomes (four telocentric and two submetacentric chromosomes) in Mehsani buffaloes. The karyotype formula of Mehsani buffaloes was as follows: 
           2n (diploid) 50 = Lm4 + Lsm6 + Lt8 + Mt8 + St22 + sex chromosomes.
